Yeast Surface Display (YSD) is a eukaryotic expression system that enables the presentation of recombinant proteins on the surface of yeast cells. This technology provides a direct link between genotype and phenotype, allowing precise screening and selection of high-affinity binders.

Yeast Surface Display Mechanism

Our YSD platform is based on the α-agglutinin display system. In this system, the antibody fragments or peptides gene sequence is fused to the Aga2p subunit, which is secreted and anchored to the yeast cell wall via disulfide bonds to the Aga1p subunit. Upon induction with galactose, the fusion protein is expressed and displayed on the yeast cell surface, enabling direct interaction with fluorescently labeled antigens during screening. This design allows efficient display of functional VHH molecules for downstream screening and affinity analysis.
